Abstract
Over the past few years, we have at the Danish Regenerative Engineering Alliance for Medicine (DREAM, www.dream.au.dk) focused on developing precision gene editing technologies and multi-OMICS technologies to understand and combat degenerative diseases. We have developed a panel of CRISPR-based genetic manipulation tools: including single/multiplexed gene knockout, gene activation, gene interference, epigenetic modifications, and even for the generation of extra chromosomal circular DNAs. Combining with next generation sequencing (NGS) technologies, and particularly single cell RNA and DNA sequencing, we have highlighted the power of all these forefront NGS technologies in decoding the complexity of cell heterogeneity normal/tumor endothelial cells, mapping the first mammalian brain transcriptome Atlas, revealing the brain-wide RNA editing events in pigs etc. One reason for our massive efforts in studying the endothelial cell heterogeneity and tissue-specific transcriptome in pigs is for better understanding genetic and molecular mechanisms underlying the xenograft immune rejection. Based on the precision genetic manipulation of the pig genome and handmade cloning, we aim for generating better humanized pigs for generative medicine.

The talk is 45 min in duration followed by 15 minutes of discussion.
