Abstract
Alzheimer’s disease (AD) is the most common form of age-related dementia affecting millions of patients worldwide. Disturbingly, disorders of the metabolism, such as obesity, diabetes, and dyslipidemia emerge as major risk factors of AD, causally linking two health problems of epidemic proportions. Still, little is known about the principles whereby metabolites control brain development and function, and how disturbances in energy balance cause neurodegeneration. Previous work has identified VPS10P domain receptors as dual regulators of systemic metabolism and brain function, and as common risk genes in metabolic and neurodegenerative diseases. Combining studies in patients and in humanized mouse and iPSC models, we elucidate molecular mechanisms of receptor (dys)function in metabolism and brain health. In this lecture, I will discuss our latest work on the roles of VPS10P domain receptors sortilin and SORLA in control of systemic and brain lipid metabolism that may provide explanatory models for the association of dyslipidemia and obesity with AD.
