Abstract

Although currently available antidepressants are effective, they all have a substantial time lag to induce a significant therapeutic response. Additionally, approximately 40% of the patients do not reach remission after several weeks of treatment. Therefore, the development of drugs that address these limitations is critical to improving public health.

Clinical and preclinical research published in the last decade has consistently shown that drugs that modulate glutamatergic signaling in the brain are potential fast-acting antidepressants. However, they induce side-effects that limit their use. The discovery of new drugs that indirectly modulate glutamatergic signaling, acting as rapid antidepressants without inducing significant side effects, is therefore of great importance. In this scenario, purinergic, nitrergic and endocannabinoid signaling can be of potential interest since they modulate glutamate release in import limbic brain regions related to depression neurobiology. In this talk, I will present some of the newest evidence from our group that supports the search for better antidepressants by targeting atypical neurotransmitters (NO and endocannabinoids) and purines.
