On Friday 11 February at 14:00, Line Stensig Lynggaard defends her PhD dissertation entitled "Asparaginase treatment in childhood acute lymphoblastic leukemia: a balance between outcome and toxicity".

In this PhD project the balance between survival and toxicity within treatment of acute lymphoblastic leukemia (ALL) with a focus on asparaginase was investigated.

Historically, survival of ALL has increased substantially, and intensification of asparaginase therapy has been fundamental in this improvement. However, asparaginase is associated with a considerable toxicity burden, and focus has to a high degree been directed toward unintended effects of therapy. Asparaginase-associated toxicity leads to short- and long-term morbidity, but even more important is that treatment-related mortality and discontinuation of scheduled therapy are challenging further improvement of survival.

The best asparaginase dosing strategy to achieve the optimal treatment efficacy while minimizing toxicity remains controversial. Thus, the studies included in this thesis aim to illuminate this critical balance between survival and toxicities within asparaginase treatment from different angles and by addressing several major factors that might affect the balance.
