The acute phase serum protein haptoglobin and the macrophage scavenger receptor CD163 work in concert to clear hemoglobin released from red blood cells, in order to protect the vasculature from the oxidative effects of the heme iron. Upon release into circulation, hemoglobin is captured and neutralized by haptoglobin and subsequently the haptoglobin-hemoglobin complex is endocytosed by macrophages via CD163. We have previously elucidated the structural and functional role of haptoglobin, however so far, the structure of CD163 and how it interacts with haptoglobin-hemoglobin has remained unknown. At the seminar, I will present recent cryoEM data on CD163 bound to haptoglobin-hemoglobin, revealing a highly unexpected receptor structure and ligand-binding mechanism.
